The purpose of this study was to establish a polymerase chain reaction (PCR)-restriction enzyme assay for detecting the hereditary hemochromatosis (HHC) mutation, C282Y, in gestational and gestational diabetic subjects in South Florida. DNA samples from 43 gestational subjects were amplified by PCR, digested with RsaI, and analyzed by electrophoresis. An allelic frequency of 2.33%, or 4.65% heterozygosity, was observed. The assay is successful and applicable to future studies on HHC and gestational diabetes. ^

An attempt has been made in this study to screen some fish muscle enzymes to assess their potential worth in testing the degree of freshness of fish. A problem with routine enzyme activity determinations is the complexity of the method of enzyme assay. Hence, in the present study as far as possible simple assay techniques were adopted. Several species were screened to assess the possibility of employing this procedure on a large scale. It is hoped that findings of this study will lead to the development of meaningful criteria in testing the freshness of fish. This thesis has been divided into five chapters

Research using 1,8-naphthalimide derivatives has expanded rapidly in recent years owing to their cell-permeable nature, ability to target certain cellular locations and fluorescent properties. Here we describe the synthesis of three new esters of 4-hydroxy-N-propyl-1,8-naphthalimide (NAP) and the development of a simple and sensitive assay protocol to measure the activity of carboxylester hydrolases. The NAP fluorophore was esterified with short (butyrate), medium (octanoate) and long (palmitate) chain fatty acids. The esters were spectroscopically characterised and their properties investigated for their suitability as assay substrates. The esters were found to be relatively stable under the conditions of the assay and levels of spontaneous hydrolysis were negligible. Non-specific hydrolysis by proteins such as bovine serum albumin was also minimal. A simple and rapid assay methodology was developed and used to analyse a range of commercially available enzymes that included enzymes defined as lipases, esterases and phospholipases. Clear differences were observed between the enzyme classes with respect to the hydrolysis of the various chain length esters, with lipases preferentially hydrolysing the medium chain ester, whereas esterases reacted more favourably with the short ester. The assay was found to be highly sensitive with the fluorophore detectable to the low nM range. These esters provide alternate substrates from established coumarin-based fluorophores, possessing distinctly different excitation (447 nm) and emission (555 nm) optima. Absorbing at 440-450 nm also offers the flexibility of analysis by UV-visible spectrophotometry. This represents the first instance of a naphthalimide-derived compound being used to analyse these enzymes.

The presence of theta-class glutathione S-transferase (GST) in marmoset monkey liver cytosol was investigated. An anti-peptide antibody targeted against the C-terminus of rGSTT1 reacted with a single band in marmoset liver cytosol that corresponded to a molecular weight of 28 kDa. The intensity of the immunoreactive band was not affected by treatment of marmoset monkeys with 2,3,7,8-tetrachlorodibenzo-p-dioxin, phenobarbitone, rifampicin or clofibric acid. Similarly, activity towards methyl chloride (MC) was unaffected by these treatments. However, GST activity towards 1,2-epoxy3-(p- nitrophenoxy)-propane (EPNP) was increased in marmosets treated with phenobarbitone (2.6-fold) and rifampicin (2.6-fold), activity towards dichloromethane (DCM) was increased by 50% after treatment of marmosets with clofibric acid, and activity towards 1-chloro-2,4-dinitrobenzene (CDNB) was raised slightly (30-42% increases) after treatment with phenobarbitone, rifampicin or clofibric acid. Compared with humans, marmoset liver cytosol GST activity towards DCM was 18-fold higher, activity towards MC was 7 times higher and activity towards CDNB was 4 times higher. Further, EPNP activity was clearly detectable in marmoset liver cytosol samples, but was undetectable in human samples. Immunoreactive marmoset GST was partially purified by affinity chromatography using hexylglutathione-Sepharose and Orange A resin. The interaction of immunoreactive marmoset GST was similar to that found previously for rat and human GSTT1, suggesting that this protein is also a theta class GST. However, unlike rat GSTT1, the marmoset enzyme was not the major catalyst of EPNP conjugation. Instead, immunoreactivity was closely associated with activity towards MC. In conclusion, these results provide evidence for the presence of theta-class GST in the marmoset monkey orthologous to rGSTT1 and hGSTT1.

A new system has been developed to determine enzyme activities of glutathione transferase θ (GSTT1-1) based on radiometric product detection resulting from the enzymic reaction of methyl chloride with 35S-labelled glutathione. In principle, the method is universally applicable for determination of glutathione transferase activities towards a multiplicity of substrates. The method distinguishes between erythrocyte GSTT1-1 activities of human 'non-conjugators', 'low conjugators' and 'high conjugators'. Application to cytosol preparations of livers and kidneys of male and female Fischer 344 and B6C3F1 mice reveals differential GSTT1-1 activities in hepatic and renal tissues. These ought to be considered in species-specific modellings of organ toxicities of chlorinated hydrocarbons.

Dihalomethanes can produce liver tumors in mice but not in rats, and concern exists about the risk of these compounds to humans. Glutathione (GSH) conjugation of dihalomethanes has been considered to be a critical event in the bioactivation process, and risk assessment is based upon this premise; however, there is little experimental support for this view or information about the basis of genotoxicity. A plasmid vector containing rat GSH S-transferase 5-5 was transfected into the Salmonella typhimurium tester strain TA1535, which then produced active enzyme. The transfected bacteria produced base-pair revertants in the presence of ethylene dihalides or dihalomethanes, in the order CH2Br2 > CH2BrCl > CH2Cl2. However, revertants were not seen when cells were exposed to GSH, CH2Br2, and an amount of purified GSH S-transferase 5-5 (20-fold excess in amount of that expressed within the cells). HCHO, which is an end product of the reaction of GSH with dihalomethanes, also did not produce mutations. S-(1-Acetoxymethyl)GSH was prepared as an analog of the putative S-(1-halomethyl)GSH reactive intermediates. This analog did not produce revertants, consistent with the view that activation of dihalomethanes must occur within the bacteria to cause genetic damage, presenting a model to be considered in studies with mammalian cells. S-(1-Acetoxymethyl)GSH reacted with 2′-deoxyguanosine to yield a major adduct, identified as S-[1-(N2-deoxyguanosinyl)methyl]GSH. Demonstration of the activation of dihalomethanes by this mammalian GSH S-transferase theta class enzyme should be of use in evaluating the risk of these chemicals, particularly in light of reports of the polymorphic expression of a similar activity in humans.

There have been recent improvements in the clinical understanding and definition of the major types of autoimmune liver disease. However, still lacking is knowledge of their prevalence and pathogenesis. Three areas of study are in progress in our laboratory. First, in type 1 autoimmune hepatitis, the search continues to identify a liver/disease-specific autoantigenic reactant. Using hepatocyte membrane preparations, immunoblotting has underlined the problem of distinguishing, among multiple reactants, those that may be causally rather than consequentially related to hepatocellular damage. Second, in primary biliary cirrhosis (PBC), the need for population screening to ascertain prevalence and detect preclinical cases can be met by a rapid automated procedure for detection, by specific enzyme inhibition in microtitre wells, of antibody (anti-M2) to the pyruvate dehydrogenase complex E2 subunit (PDC-E2). Third, the structure of the conformational epitope within the inner lipoyl domain of PDC-E2 is being investigated by screening random phage-displayed peptide libraries using PBC sera. This has yielded phage clones in which the sequence of the peptide insert portrays the structure of this epitope, as judged by clustering of PBC-derived sequences to particular branches of a guide-tree that shows relatedness of peptides, and by reactivity of selected phage clones with anti-PDC-E2. Thus phage display identifies a peptide 'mimotope' of the antibody epitope in the inner lipoyl domain of PDC-E2.

Supramolecular chemistry is an emerging tool for devising materials that can perform specified functions. The self-assembly of facially amphiphilic bile acid molecules has been extensively utilized for the development of functional soft materials. Supramolecular hydrogels derived from the bile acid backbone act as useful templates for the intercalation of multiple components. Based on this, synthesis of gel-nanoparticle hybrid materials, photoluminescent coating materials, development of a new enzyme assay technique, etc. were achieved in the author's laboratory. The present account highlights some of these achievements.

O objetivo principal deste estudo foi investigar a interação de 24 cepas de E. faecalis isoladas de infecções endodônticas primárias às proteínas de matriz dentinária, como também a moléculas de matriz presentes em lesões de endocardites. A análise desta interação foi feita através de técnica enzimática, com confirmação pela técnica de fluorescência. Além disto, foi realizada a confirmação do isolamento da espécie E. faecalis, através da técnica de PCR para o gene 16SrRNA e a análise da presença de genes de virulência da referida espécie microbiana para aderência às supostas proteínas de matriz incluindo às de ligação ao colágeno (ace, gelE, esp, agg e efaA). O maior padrão de interação das cepas ocorreu com a fibronectina (83,4%), seguido pelo fibrinogênio (62,5%) e colágeno humano tipo I (52%). Curiosamente, a aderência observada para o colágeno do tipo I, foi de pequena magnitude, quando comparado com a amostra padrão da ATCC 29212. As cepas ATCC 29212, A1, A43 e A68 interagiram com todas as proteínas de matriz utilizadas neste estudo. Um percentual expressivo das cepas testadas apresentou amplificação para efaA (86,9%) e para ace (73,9%). Paralelamente, todas as cepas apresentaram amplificação para gelE e foram negativas para os genes agg e esp. Adicionalmente, não houve correlação entre a detecção dos genes de virulência e a interação às proteínas de matriz, evidenciando que, mesmo com a detecção dos genes nas amostras, se faz necessário avaliar a expressão gênica por qPCR.

羊草 (Leymus chinensis (Trin.) Tzvel. ) ,隶属禾本科赖草属,是欧亚大陆草原区东部的重要草原建群种之一。羊草是牧草之王,属于我国有比较优势的战略性生物资源,对我国北方畜牧业发展以及保护生态环境均具有重要作用。 羊草较弱的有性生殖特性限制了其应用,本文从实验生物学角度,研究了羊草有性生殖的基本特点,并试图通过现代分子生物学手段探讨自交不亲和性的有关机理。本论文的主要结果如下: 1. 实验发现羊草具有自交不亲和性。以6 个羊草居群为材料,测定得知开放授粉时的结实率在6.5% - 56.7%之间,自交时结实率为0.6% - 4.3%,差异极其显著; FDA 染色法检测结果显示羊草成熟花药中有活性的花粉达到92.2% 以上;在发育时间顺序和空间结构上,羊草雌蕊、雄蕊适于异花和自花授粉;花粉柱头亲和性实验表明,自交花粉只有5.5%-11.7% 是亲和的,杂交花粉亲和率达到了60.0%-84.8%,说明自交花粉在柱头上萌发受到抑制,其次,荧光显微镜还观察到“不亲和花粉”在进入柱头后生长缓慢,或停止延伸。 2. 初步确定羊草自交不亲和性具有配子体型遗传特点。以不同居群羊草杂交后的姊妹系作为实验材料,观察到自交组合的亲和率变幅为0 % - 6.9 %,杂交组合的亲和率具有连续性变异和变幅较宽的特点(47.5% - 96.0 %),且正反交结果具有一定的一致性(88.2%),表现出配子体遗传特性。 3. 羊草居群内结实率存在一定变异。以羊草单株为单位分别进行自交、随机互交和开放授粉,结果显示三者的平均结实率分别为4.6%,18.1% 和35.7%,株间的变异系数分别为33.4%,21.2%和17.1%,这些株间的变异均达到统计上的显著差异;同时羊草自交、杂交和开放授粉之间具有一定的相关性,显示羊草的这种株间差异与株系本身的生理特性相关。 4. 分离了羊草硫氧化还原蛋白 H 基因(ThioLc)并对其功能进行了分析。克隆了ThioLc全长和cDNA序列。序列分析结果显示,DNA全长2257 bp,包括3 个内含子和4 个外显子,与水稻Thio h 的cDNA 序列相比,具有 32.0% 的同源性;Southern 杂交显示 ThioLc 在羊草基因组中是单拷贝;Northern 杂交显示 ThioLc 在羊草根、茎、叶和幼小的雌蕊中没有表达, 在成熟雌蕊和幼小的花粉中微量表达, 在成熟花粉中大量表达,说明分离的羊草硫氧化还原蛋白H 基因具有花粉特异表达特点。 5. 原核表达的ThioLc 蛋白具有较高的催化活性。构建了ThioLc 基因的原核表达载体,检测证明ThioLc基因在大肠杆菌中正常表达;提取表达蛋白,纯化,用胰岛素和二硫苏糖醇反应体系进行硫氧化还原蛋白的催化作用反应,结果表明表达的蛋白具有催化活性。这一结果为进一步搜寻靶向蛋白和研究该蛋白的结构、功能和作用方式奠定了基础。

A new enzyme assay method for screening alpha-glucosidase inhibitors with rapidity and simplicity was developed. The enzyme-substituted alpha-glucosidases for this assay was glucoamylase. Samples were spotted or developed on the silica gel plate. The agar solution containing substrate was poured on the plate, and paper impregnated with enzyme was layered on the agar. After incubation, an inhibitory circle would appear around the inhibitor. By using this method, more than 200 strains of marine microorganisms were screened. Among them, three active strains were found to secrete inhibitors in the culture medium.

Manganese superoxide dismutase (MnSOD) plays an important role in crustacean immune defense reaction by eliminating oxidative stress. Knowledge on MnSOD at molecular level allows us to understand its regulatory mechanism in crustacean immune system. A novel mitochondrial manganese superoxide dismutase (mMnSOD) was cloned from hepatopancreas of Chinese shrimp Fenneropenaeus chinensis by 3' and 5' rapid amplification of cDNA ends (RACE) PCR. The full-length cDNA consists of 1185 bp with a 660 bp open reading frame, encoding 220 amino acids. The deduced amino acid sequence contains a putative signal peptide of 20 amino acids. Sequence comparison showed that the mMnSOD of F. chinensis shares 88% and 82% identity with that of giant freshwater prawn Macrobrachium rosenbergii and blue crab Callinectes sapidus, respectively. mMnSOD transcripts were detected in hepatopancreas, hemocytes, lymphoid organ, intestine, ovary, muscle and gill by Northern blotting. RT-PCR analysis indicated that mMnSOD showed different expression profiles in shrimp hemocytes and hepatopancreas after artificial infection with while spot syndrome virus (WSSV). In addition, a fusion protein containing mMnSOD was produced in vitro. LC-ESI-MS analysis showed that two peptide fragments (-GDVNTVISLAPALK- and -NVRPDYVNAIWK-) of the recombinant protein were identical to the corresponding sequence of M. rosenbergii mMnSOD, and the enzyme activity of the refolded recombinant protein was also measured.
